Molecular Identification and FTIR Analysis of Polythene Degrading Bacteria Isolated from Refuse Dump Soil

Abstract

Low-density polyethylene (LDPE) is one the most widely used packaging material after cardboard,primarily because of its convenience and low cost. Its increasing accumulation in the environment hasbecome a threat to the planet. However, due to its ubiquitous and recalcitrant nature to degradation,disposal strategies are crucial and need attention. Recently, microorganisms have become the focus ofinterest for environmentally friendly disposal of plastic waste. This research aimed to investigate thepolythene biodegrading potency of bacteria isolated from three different refuse dumpsites in Lapai, NigerState, Nigeria. A total of 13 bacterial isolates were recorded with a maximum percentage weight loss of34% by 2G after incubation of polyethylene with isolated bacteria at 37°C for 6 weeks. The rate ofbiodegradation was further confirmed by FTIR analysis which indicated changes in some bonds withinthe treated samples as compared to the control. Analysis of the 16S rDNA sequence of 2G showed that ithad 87% pairwise similarity with Proteus mirabilis Xuyi_408_2. Hence, Proteus mirabilis, Xuyi_408_2as identified by the 16s ribosomal RNA (rRNA) gene sequence homology with molecular weight of1.5kbp, is the most efficient polyethylene degrading bacteria as compared to other isolates.
